Oocyte formation by mitotically active germ cells

Germline stem cells that produce oocytes in vitro and fertilization-competent eggs in vivo have been identified in and isolated from adult mouse ovaries, Nature Medicine reported on March 26.

The magazine describes and validates a fluorescence-activated cell sorting-based protocol that can be used with adult mouse ovaries and human ovarian cortical tissue to purify rare mitotically active cells that have a gene expression profile that is consistent with primitive germ cells.

Once established in vitro, these cells can be expanded for months and can spontaneously generate 35- to 50-μm oocytes, as determined by morphology, gene expression and haploid (1n) status, the report said..

Injection of the human germline cells, engineered to stably express GFP, into human ovarian cortical biopsies leads to formation of follicles containing GFP-positive oocytes 1–2 weeks after xenotransplantation into immunodeficient female mice, it said.

Thus, ovaries of reproductive-age women, similar to adult mice, possess rare mitotically active germ cells that can be propagated in vitro as well as generate oocytes in vitro and in vivo, the magazine said.

미국 매사추세츠종합병원 연구진은 여성의 난소에서 채취한 난소줄기세포로 난자를 만들었다고 지난달 26일(현지 시간) 과학학술지 <네이처 메디신(Nature Medicine)>에 발표했다.

이들은 줄기세포를 분화해 얻은 난모세포를 초록색으로 염색한 다음 쥐의 난소에 넣어 관찰한 결과 14일 뒤 성숙한 난자로 자라난 것을 확인했다고 밝혔다. 난소는 일본에서 성전환수술을 받은 20대 여성에게서 기증받았다.

난소는 크게 두 종류의 세포로 이뤄진다. 난자를 만드는 난원세포와 난소를 이루는 일반 체세포다. 지금까지 난자와 정자는 각각의 씨앗이 되는 난원세포와 정원세포에서만 만들어진다고 여겼다. 현재 생물학 교과서도 그렇게 돼 있다.

그런데 이번 실험에서 연구진은 난소줄기세포를 체세포에서 얻었다. 난원세포가 아닌 난소를 구성하는 체세포로도 난자를 만들 수 있다는 게 증명된 셈이다.

임정묵 서울대 농생명과학부 교수는 "교과서 내용이 바뀔 만한 새로운 발견"이라며 "조기폐경으로 난자를 만들 수 없는 여성 등의 불임을 극복하는데 향후 큰 도움이 될 것"이라고 말했다.

강경선 서울대 수의학과 교수는 "난자를 사용한 게 아니라 난소에 있는 체세포를 쓴 것이어서 윤리적인 문제도 없다"고 했다.

이번 연구를 이끈 조너선 틸리 박사는 이미 2004년 비슷한 연구 결과를 발표한 적이 있다. 그는 과학학술지 <셀(Cell)>에 난소의 체세포에서도 줄기세포를 얻을 수 있다는 연구 결과를 냈다. 그러나 학계는 이를 받아들이지 않았고, 그가 몸담고 있는 미국 하버드대에선 사기꾼으로 몰리기도 했다. 2004년 발견한 그 가능성을 실험으로 확인, 인정받기까지 8년이 걸린 것이다.
