North Korea threatened to confiscate all real estate on Mt. Geumgang owned by South Korean companies unless its owners show up for an inspection on March 25.
The Ministry of Unification expressed deep regret over the announcement.
Besides Hyundai Asan, operator of the Mt. Geumgang tour program, some other companies own property at the resort, such as hotels, restaurants and shops.
The exact number of the owners was unavailable, according to the ministry. The amount of investment is over 359 billion won.
The South Korean government also owns one building, for which it paid 60 billion won.
Pyongyang renewed its threat to scrap the tourism accord with Seoul unless the South lifts its embargo on tour programs to North Korea by the end of this month.
The North's Asia-Pacific Peace Committee, the state body in charge of inter-Korean exchange, announced it would pick a third tour operator in North Korea or China, unless Hyundai Asan resumes sending tourists from April 1.
The tour program to Mt. Geumgang came to a halt two years ago after a South Korean woman tourist was shot to death. The Gaeseong tour was also suspended later.
The South has blocked tours of civilian tourists over safety concerns, which angered the North.
"There's no change in our stance that without a security assurance, the South Korean government won't resume the tour programs to Mt. Geumgang and Gaeseong," the ministry said.
Hyundai Asan did not make a public response to the North's announcement. But the notice added chaos to what's already a complicated situation. Cho Kun-shik, chief of Hyundai Asan, announced his resignation Thursday.
Since it began in 1998, the Mt. Geumgang tours have generated more than $480 million in fees for the North. Cross-border visitors could previously take day trips to the city of Gaeseong across the border.
The city is also the location of a joint industrial zone, where 42,000 North Koreans work in 110 South Korean factories.

통일부에 따르면 부동산 소유주의 정확한 인원은 입수 가능하지 않다. 투자액은 3,590억 원이 넘는 것으로 알려졌다.
남한 정부도 600억을 투입한 건물 한 채를 소유하고 있다.
북측은 이 달 말까지 북한 관광에 대한 제재를 남측이 풀지 않으면 양측이 체결한 관광 합의안을 폐기하겠다고 되풀이했다.
남북 교류를 담당하고 있는 국가 조직인 북한의 아태평화위원회는 현대아산이 4월 1일부터 남한 사람들의 북한 관광을 재개하지 않으면 자국이나 중국에서 제3의 운영자로 바꾸겠다고 발표했다.
금강산 관광 사업은 남측 여성 관광객 한 명이 사살된 2년 전 중지됐다. 뒤이어 개성 관광 역시 중단됐다.
남측은 안전문제를 이유로 일반 관광객들의 북한 관광을 봉쇄해 북측의 분노를 샀다.
